Isolation, characterization, and complete genome analysis of P1312, a thermostable bacteriophage that infects Thermobifida fusca

摘要: Thermobifida fusca is a moderately thermophilic and cellulolytic actinobacterium. It of particular interest due to its ability not only produce variety biotechnologically relevant enzymes but also serve as an alternative host for metabolic engineering the production valuable chemicals from lignocellulosic agricultural wastes. No bacteriophage that infects T. has been reported, despite potential impacts on utilization fusca. In this study, extremely thermostable P1312 was isolated manure compost. Electron microscopy showed icosahedral head long flexible non-contractile tail, characteristic family Siphoviridae. double-stranded DNA genome 60,284 bp with 93 ORFs. Thirty-one ORFs encode proteins having putative biological functions. The genes involved in phage particle formation cluster together region approximately 16 kb, followed by segment containing presumably degradation/modification cell wall disruption. required replication transcriptional control are dispersed within rest genome. Phylogenetic analysis large terminase subunit suggests headful packaging chromosome circularly permuted direct terminal repeats.
