摘要: A simple and reliable method was established for the maintenance of a permanent stock several Medicago truncatula genotypes selected from general seed by their in vitro culture amenability embryogenic capacity. In first step, multiple shoots were induced cotyledon axillary meristem pre-germinated seeds Murashige Skoog medium supplemented with cytokinins (9.3 μM zeatin, 22.2 benzylaminopurine or 4.5 thidiazuron). second allowed to develop growth-regulator-free medium. Benzylaminopurine at supported best combination shoot quality number produced. Rooting microshoots depended on cytokinin used induction faster zeatin-treated shoots. this work propagation system devised where addition growth regulators restricted phase therefore reducing risks epigenetic somaclonal variation.
