Prokaryotic expression of chicken interferon-γ fusion protein and its effect on expression of poultry heat shock protein 70 under heat stress

摘要: Interferons have attracted considerable attention due to their vital roles in the host immune response and low induction of antibiotic resistance. In this study, total RNA was extracted from spleen cells chicken embryos inoculated with Newcastle disease vaccine, full-length interferon-γ (ChIFN-γ) gene amplified by RT-PCR. The full complementary DNA sequence ChIFN-γ 495 bp long cloned into prokaryotic expression vector pProEX™HTb. plasmid transformed Escherichia coli DH5α induced isopropyl β-D-1-thiogalactopyranoside. Sodium dodecyl sulfate – polyacrylamide gel electrophoresis Western blot results showed expressed fusion protein had a molecular weight approximately 18 kDa recognized an anti-His mAb. Moreover, found demonstrate anti-viral activity vitro. To test vivo function broilers under heat stress, 100 were randomly assigned either control group or treated group, which they hypodermically injected recombinant ChIFN-γ. Results demonstrated affects messenger levels shock 70 (HSP70) heart lung tissues, decreases concentration HSP70 serum. Therefore, we conclude can reduce stress some extent vivo.
