The conversion of RFLP markers to allele specific amplicons linked to QTLs governing malting quality in barley

摘要: Application of marker-assisted selection with RFLP based markers has been constrained by high cost and time requirements in situations involving a large number plants. mapped on Harrington/TR306 population have identified elsewhere as linked to quantitative trait loci (QTL) governing malting quality. The probes ABG610, ABC622, well for the Nar1, Amy1 Nar7 were sequenced locus specific primers developed. These applied genomic DNA from both Harrington TR306. Sequence analysis resultant monomorphic fragments revealed sequence divergence Xabg610, Xabc622, Nar1 loci, but not locus. set Hor2 barley lines TR306 led direct amplification codominant alleles. Allele-specific designed among Xabc622 Amplification conditions optimized each these alleles such that only favourable allele was amplified. usefulness selecting demonstrated their failure amplify corresponding lines, Sterling, Stella WM872. allele-specific amplicon capable differentiating this between conversion into PCR amplifiable, amplicons would greatly facilitate application breeding programs.
