摘要: Cucumber mosaic virus (CMV) is an important pathogen of lupins in Australia which causes serious yield losses up to 60% epidemic years. In commercially grown lupin (Lupinus angustifolius and L. luteus) crops CMV spread non-persistently by aphid vectors, but it can also be seed borne this extends infection into successive generations. Resistance has been identified luteus cv. Wodjil the conferred Ncm-1 gene. The aims research were study gene order gain a better understanding resistance yellow lupin, develop molecular marker linked for use assisted selection. Previously published data Jones et al (1996) as being single dominant gene, however, phenotypic analysis segregating mapping population thesis was consistent with modified at least one other minor polygenic nature genetic background further supported AFLP major QTL associated resistance. A PCR based approach, using degenerate primers designed on conserved disease protein motifs, used identify analogues (RGA) luteus. Comparative revealed that RGAs isolated from members TIR-NBS-LRR class R proteins similar TMV N tobacco RT4-4 pepper. Extensive comparative genomes model species (including Medicago truncatula, Glycine max, Arabidopsis thaliana Lotus japonicus) explored validated assignment category candidate resistance. found highly both resistant susceptible varieties tested. SNPs resulted non-synonymous mutations cDNA 5’ RACE nucleotide primer extension (SNuPE) assays MALDI-ToF mass spectrophotometric analysis. As SNuPE allele specific nucleotide, genotyping accurate provides co-dominant information. Two developed bulked samples two L.luteus populations One assay, A267→C associate This assay first its kind reported lupin.
