The detection of natural opal suppressor seryl-tRNA in Escherichia coli by the dot blot hybridization and its phosphorylation by a tRNA kinase

摘要: Abstract It was believed that there no natural suppressor tRNA in Escherichia coli, however, it has been suggested selC, relating to the synthesis of formate dehydrogenase a selenoprotein [(1988) Nature 331, 723–725], codes for tRNA, even though presence not yet demonstrated. We detected product selC preparation E. coli MC 4100 strain by dot blot hybridization method with DNA probe (ACCGCTGGCGGC) corresponding extra arm tRNA. Two peaks were found chromatographic pattern from Sephadex A50. The amount estimated be about 0.03% total [3H]seryl-tRNA phosphorylated kinase B. These results suggest opal seryl-tRNA should converted selenocysteyl-tRNA through phosphoseryl-tRNA, and occurs vertebrates as general phenomenon.
