摘要: Background  The intimate association between glial cells and neurons within the enteric nervous system has confounded careful examination of direct responsiveness glia to different neuroligands. Therefore, we aimed investigate whether neurotransmitters known elicit fast excitatory potentials in nerves also activate directly. Methods  We studied effect acetylcholine (ACh), serotonin (5-HT), adenosine triphosphate (ATP) on intracellular Ca2+ signaling using aequorin-expressing Fluo-4 AM-loaded CRL-2690 rat human cell cultures devoid neurons. influence these proliferation was measured their expression c-Fos as well fibrillary acidic protein (GFAP), Sox10, S100 examined by immunohistochemistry quantitative RT-PCR. Key Results  Apart from ATP, ACh 5-HT induced a dose-dependent increase Ca2+ concentration cells. Similarly, evoked Ca2+ transients primary obtained mucosal biopsies. In contrast with stimulation early gene GFAP, were not affected following neurotransmitters. Conclusions & Inferences  provide evidence that respond changes Ca2+. On basis our experimental vitro setting, show are only directly responsive purinergic but serotonergic cholinergic signaling mechanisms.
