Exploiting lectin affinity chromatography in clinical diagnosis.

摘要: Lectin affinity chromatography (LAC) offers a tool that aids purification of cell surface glycoconjugates in sufficient quantities so studies addressing their structural elucidation could be carried out. It has several advantages over the conventional biochemical methods, such as immunoprecipitation and/or immunoaffinity chromatography, used for various glycoconjugates. Serial LAC (SLAC) not only helps establish identity glycoprotein or allows to homogeneity from among mixture glycoproteins, but it also successfully resolves microheterogeneity these which is an otherwise impracticable problem address. Specific cases altered expression and maintenance some glycoproteins pathological conditions vis during normal biology are presented. The application (i) itself, (ii) serial fashion, (iii) conjunction with other techniques two-dimensional electrophoresis, capillary mass spectrometry, etc. diagnosis certain conditions, possibility using this knowledge designing treatments diseases, discussed.
