Effects of Ethanol Infusion on the Redox State and Metabolite Levels in Rat Liver in vivo

摘要: 1 Hepatic levels of several metabolites the Embden-Meyerhof pathway, citric acid cycle and oxidized reduced substrate pairs lactate, α-glycerophosphate, β-hydroxybutyrate, malate, glutamate dehydrogenase systems were measured in freeze clamped livers from fed male rats, before after ethanol infusion. 2 Ethanol infusion rats 30 min resulted an increase hexosemonophosphates, triosephosphates, fructose 1,6 diphosphate, glutamate. [Lactate]/[pyruvate] ratio increased about 3 fold, [α-glycerophosphate]/[dihydroxyacetonephosphate] 1.5 [malate]/[oxaloacetate], [β-hydroxybutyrate]/[acetoacetate], [glutamate]/[2-oxoglutarate] × [NH4+] 2 fold. 3 The pyruvate, acetyl CoA, citrate, ATP decreased during treatment. 4 The hepatic glycogen, oxaloacetate, 2-oxoglutarate, ammonia, aspartate, acetoacetate remained unchanged, infusion, as compared to values infusion. 5 Hepatic glucose 6-phosphatase activity was significantly 15 (1.5 g/kg body weight). The showed a maximum diphosphatase did not change infusion. Hepatic alkaline phosphatase activities with β-glycerophosphate no significant changes normal rat livers. 6 The presence (8 mM) incubation medium effect on [6-14C]glucose incorporation into glycogen by liver slices.
