摘要: Polyphenol oxidase (EC 1.10.3.1) was extracted from an acetone powder prepared Koshu grapes. The enzyme purified 172-fold by ammonium sulfate fractionation followed three successive column chromatographies on O -(carboxymethyl) cellulose, -(diethylaminoethyl) cellulose and hydroxylapatite. homogeneous polyacrylamide gel electrophoresis. molecular weight of this estimated to be 39 000 - 41 filtration sodium dodecyl electrophoresis, the molecule had no subunit. temperature pH optima were 25°C 6.0, respectively. stable in alkaline range (between 7.0 11.0) at temperatures up 30°C. exhibited high activity toward o -diphenolic compounds showed monophenols.
