Application of iTRAQ quantitative proteomics in identification of serum biomarkers in breast cancer

摘要: Aim: This study aims to explore the presence of informative protein biomarkers in human serum proteome. Patients and Methods; Serum samples collected from 20 breast cancer patients which were divided into two groups cancer, 10 health controls mammary fibroma profiled using iTRAQ technology coupled with LC-ESI-QTOF-MS, Mascot searching. Western-blotting used for validation candidate on a new group healthy subjects as well patients. Results: 335 proteins identified, 11 associated includind 1 upregulated downregulated proteins. Three biomarker candidates generated experiments successfully verified Western-blotting. Conclusion: provided global view potential mechanisms potentional demonstrated that combined LC-ESI-QTOF-MS quantitative proteomics is powerful tool discovery.
