摘要: The distribution of ribonucleoprotein (RNP) within the mitotic spindle newt lung epithelial cells was studied with high voltage electron microscope (HVEM) using Bernhard's uranyl-EDTA-lead staining thick sections in conjunction ribonuclease digestion fixed cells. results indicate that aside from ribosomes, major RNP-containing components are kinetochores and centrioles, both which stain electron-opaque after EDTA treatment. In cases, material associated these microtubule organizing centers (MTOC's) can be removed by RNAse cold perchloric acid (PCA) extraction under conditions leave microtubules (Mts) intact. reaction is not abolished PCA alone or substituting other positively charged proteins (i.e., cytochrome c lysozyme) for RNAse. RNP component kinetochore closely bases microtubules. centriole seen to surround triplet blades. No evidence found presence pericentriolar material. possible function centriolar discussed.
