Cell Origin and Culture History Determine Successful Integration of Neural Precursor Transplants into the Dentate Gyrus of the Adult Rat

摘要: The success of transplants neural tissue into the adult dentate gyrus in generating mature neurons is highly variable. Here we address roles origin and its pre-implantation preparation, show that both are critical. We transplanted neonatal cultured or primary rat cells from either ventral subventricular zone (vSVZ) (DG) DG. Only DG robustly generated (80% NeuN Prox1-positive at 6 weeks), substantially repaired damaged DG, formed glutamatergic projections to target CA3 region. Cultured expanded for 7 days showed limited neuronal differentiation after transplantation (10% cells) whereas vSVZ failed make any granular neurons. found a specific population postmitotic young (triple doublecortin/NeuN/Prox1-positive) were particularly abundant cells, but markedly reduced absent cells. Labelling with mitotic marker BrdU suggested source in-vivo. conclude pre-transplantation history donor key factors determine outcome transplantation. These findings may be therapeutic interest cell replacement therapy treating hippocampus.
