摘要: A radioimmunoassay, selective for the clinically important mid region of human parathyroid hormone (hPTH), is reported. The synthetic44–68 amino acid sequence (h44–68PTH) was used as both standard and tracer material. This eliminated many undesirable characteristics associated with PTH assays that employ biological origin. These components allowed detection regional fragments present in intact fragmented hPTH, a working range between 50 2500 pg/ml h44–68PTH. There no interference from serum proteins significant cross reactivity to N-terminal, C-terminal other hPTH peptides. assay proved be rapid (total time 24 h) extremely reproducible, an intraassay interassay variation 2.8% 5.6% respectively (expressed percentage SE mean). plasma concentration normal subjects established 129 ± 6 50–300 (n = 42). assay, using fully synthetic peptides, able distinguish euparathyroid hyperparathyroid subjects, which suggests diagnostic value.
