摘要: Sample preparation, including bacterial lysis, remains a hurdle in the realization of complete point-of-care tests for many pathogens. Here, we developed sample preparation methodology enzymatic lysis and heating low-resource, applications. We show an instrument-free chemical heater system rapid Gram-positive bacterium (Staphylococcus aureus) RNA virus (human respiratory syncytial virus) using dried enzyme mixture (achromopeptidase) S. aureus. After step (<1 minute), enzymes are heat deactivated (<5 minutes) simple disposable heater. demonstrated that both DNA heat-treated could be directly amplified without purification, even presence clinically-obtained human nasal sample. This approach to dry storage is adaptable applications where samples need lysed, use low-resource laboratories single-use or cartridge-based diagnostic devices.
