摘要: Infarct size delineation by triphenyltetrazolium chloride (TTC) staining is dependent on sufficient reperfusion. We therefore evaluated the possibility of using propidium iodide (PI), a reagent conventionally used in flow cytometry to fluorescently stain dead cells, for infarct analysis after short periods Forty-five rabbits were subjected either 15 min, 2 h or 4.5 coronary artery occlusion without reperfusion, 30 min and followed 1 3 Fifteen before terminating experiment, PI was injected into left atrium. Patent blue violet delineate area at risk. Following incubation TTC, risk excised cross sections obtained microscopical quantification fluorescence. fluorescence absent permanent control areas. sizes measured TTC significantly smaller reperfusion as compared (30 occlusion: 1+/-1 v 34+/-9%; P<0.05; 9+/-6 47+/-8%; P<0.01). In contrast, determined reached values comparable those conventional histology longer times already 35+/-16.5%; 61+/-8%). Therefore, measurement more reliable than staining.
