Injectable PAMAM/ODex double-crosslinked hydrogels with high mechanical strength

摘要: In situ injectable double-crosslinked hydrogels containing thiol functionalized poly(amido-amine) dendrimers (Gn-PAMAM-NH2-X) and oxidized dextrans (ODex) were prepared under physiological conditions without using potentially cytotoxic cross-linkers. The structure was created by Schiff's base reaction the formation of disulfide bonds. morphology characterized scanning electron microscopy. gelation time, swelling rheological behaviors investigated. We also studied adhesive strength cytocompatibility hydrogels. surface amino density, concentration generation PAMAM are main factors affecting gelation. Relatively high density contributes to quick gelation, whereas too great a may lead brittleness hydrogel. A moderate (10% wt) is suitable for considering its appropriate time. Where mass PAMAM-NH2 identical, with less prone PAMAM/ODex have structures crosslinking which their storage modulus. about 2.4 times commercial available fibrin glue these nontoxic L929 mouse fibroblast cells. cells can attach easily proliferate on them, demonstrates novel biocompatible potential uses in tissue engineering.
