摘要: More than 99% of microorganisms on the earth are unculturable with known culturing techniques. The emergence metagenomics high-throughput sequencing technologies has enabled researchers to capture a comprehensive view complex bacterial community which comprises both culturable and species. However, function an individual species remains difficult elucidate in conventional metagenomic study, generates numerous genomic fragments unidentifiable origins at or genus level. This limitation hampers any in-depth investigations its members. Recently, as alternative compensatory approach, genomics targeting single been proposed. In this whole-genome amplification technique using Phi29 DNA polymerase is applied obtain sufficient quantity for genome sequence analysis from only thousand cells. It expected that combination single-species-targeting provides great progress understanding ecology, physiology, evolution microbial communities.
