TET1 is a negative transcriptional regulator of IL-1β in the THP-1 cell line

摘要: TET1 is a member of the recently identified family epigenetic regulators, TET1-3 which catalyze enzymatic conversion methyl mark on cytosine (methylcytosine, mC) to hydroxymethyl (hmC). The functions hmC are required for stem cell maintenance and controlling differentiation reprogramming. So far, no roles TET proteins have been in cells immune system. Here we show that negative regulator IL-1β transcription following an inflammatory stimulus negatively modulates secretion THP-1 cells. In addition, expression regulated during inflammation both primary dendritic Importantly, other highly induced pro-inflammatory genes also by TET1, including cytokines, chemokines adhesion molecules. with known regulation, TET2, may participate according our observations. Our results suggest TET1-dependent anti-inflammatory pathway, include TET2. particular, transcriptional regulation likely depend TET1-regulated chromatin domains. This work highlights contribution mechanisms efficient organization responses.
