摘要: The mi locus of mice encodes a transcription factor the basic-helix-loop-helix-leucine zipper protein family (MITF). MITF encoded by mutant allele ( -MITF) deletes 1 4 consecutive arginines in basic domain. mi/mi genotype express -MITF, whereas tg/tg have transgene at 5′ flanking region gene and do not any MITF. To investigate function -MITF cultured mast cells (CMCs), we took two approaches. First, mRNA obtained from CMCs or was subtracted complementary (c) DNA library normal (+/+) CMCs, (+/+- ) subtraction libraries were obtained. When number clones that hybridized more efficiently with +/+ CMC cDNA probe than compared using Southern analysis, larger library. Second, expression six genes between Northern analysis. three encoding mouse cell proteases impaired both CMCs.
