摘要: Osteosarcoma therapy might be moving toward nanotechnology-based drug delivery systems to reduce the cytotoxicity of antineoplastic drugs and improve their pharmacokinetics. In this paper, we present, for first time, an extensive chemical in vitro characterization dual-loaded photo- chemo-active keratin nanoparticles as a novel system treat osteosarcoma. The are prepared from high molecular weight hydrosoluble keratin, suitably functionalized with photosensitizer Chlorin-e6 (Ce6) then loaded chemotherapeutic Paclitaxel (PTX). This multi-modal PTX-Ce6@Ker nanoformulation is by both drug-induced aggregation desolvation methods, comprehensive physicochemical performed. efficacy tested on osteosarcoma tumor cell lines, including chemo-resistant cells, using 2D 3D model systems. single combined contributions PTX Ce6 evaluated, results show that retains its activity while being vehiculated through keratin. Moreover, act additive manner, demonstrating combination cytostatic blockage oxidative damage ROS upon light irradiation have far superior effect compared singularly administered or Ce6. Our findings provide proof principle development novel, treatment
