Analysis of the myosin-II-responsive focal adhesion proteome reveals a role for β-Pix in negative regulation of focal adhesion maturation.

摘要: Focal adhesions undergo myosin-II-mediated maturation wherein they grow and change composition to modulate integrin signalling for cell migration, growth differentiation. To determine how focal adhesion is affected by myosin II activity, we performed proteomic analysis of isolated compared protein abundance in from cells with without inhibition. We identified 905 proteins, 459 which changed inhibition, defining the myosin-II-responsive proteome. The 73% proteins proteome was enhanced contractility, including involved Rho-mediated endocytosis- calpain-dependent disassembly. During 27% proteome, Rac-mediated lamellipodial protrusion, were enriched adhesions, establishing that recruitment also negatively regulated contractility. focused on Rac guanine nucleotide exchange factor β-Pix, documenting its role negative regulation promotion protrusion turnover drive migration.
