摘要: The regulatory control of human erythropoiesis through a purified protein, activin A, was examined. Previous studies using mixed populations bone marrow cells suggested that A has an indirect effect on cellular proliferation and DNA synthesis erythroid progenitors the mediation accessory cells. In present studies, cultures were used to examine globin gene expression. Human burst-forming units (BFU-E) partially from peripheral blood, after 8 days culture generated consisted mainly colony-forming (CFU-E). It found subsequent 7-day these yielded similar numbers size distributions colonies, regardless presence in cultures. addition, progenitor responsive, terms stimulation synthesis, addition erythropoietin, but not treatment by A. Therefore, once are depleted cells, little both progenitors. However, when cultured levels all alpha, beta, epsilon transcripts hemoglobins significantly increased. disuccinimidyl suberate chemically cross-link 125I-activin cell surface binding proteins (45 54 Kd) K562 labeling specifically inhibited unlabeled transforming growth factor-beta. These results suggest that, its progenitors, directly affects mRNAs developing specific receptor(s).
