摘要: Circulating erythropoietic precursors in normal men and patients with hemoglobinopathies were characterized culture. Blood mononuclear cells harvested a modification of the Ficoll-Isopaque technique cultured methylcellulose for 14 days. The majority colonies consisted several subcolonies assuming morphology “bursts” described murine marrow cultures. Time course studied colony formation from blood nucleated confirmed that circulating represented only early stages development. Peak sedimentation velocity analyzed using Staput apparatus averaged 5.31 mm/hr corresponded human marrow. One ml yielded an average 153 785 hemoglobinopathies. No correlation was observed between reticulocyte indices individual patients. Examination proliferative state high specific activity tritium-labeled thymidine revealed almost none or DNA synthetic phase.
