Microsatellite alterations in various sarcomas in Japanese patients.

摘要: To determine the usefulness of microsatellite analysis in differential diagnosis various sarcomas, we investigated alterations at 12 loci by polymerase chain reaction and electrophoresis 39 Japanese patients with sarcomas. The sarcomas were: osteosarcoma, Ewing's sarcoma, chondrosarcoma, liposarcoma, leiomyosarcoma, epithelioid rhabdomyosarcoma, synovial malignant fibrous histiocytoma. We also examined ten leiomyomas to contrast leiomyosarcoma. No instability (MSI) or loss heterozygosity (LOH) were found histiocytoma, leiomyoma. Only three patients, one each manifested MSI, whereas, sarcoma LOH, an incidence 43%, 14%, 86%, 20%, 75%, respectively. Interestingly, showed unusual patterns probably due intratumoral heterogeneity. Kaplan-Meier revealed that LOH on 11p was predictive poor prognosis osteosarcoma. low MSI indicates is not necessary for neoplastic transformation However, very high leiomyosarcoma may serve versus Microsatellite predict
