摘要: The in vitro chemosensitivity of 11 human colorectal cell lines to seven chemotherapeutic agents was determined using a semiautomated tetrazolium-based colorimetric assay (MTT assay). Four the were from primary tumors and metastases. Eight patients with no prior chemotherapy. From results, we predict 5-fluorouracil (5-FU) be sole active agent tested. This is based on two observations: range drug concentrations which produced 50% inhibition growth greatest 5-FU (388-fold versus 5- 30-fold other six agents); area under curve (AUC) within clinically achievable only for 5-FU. Since AUC at lines, performed multivariate analysis explore parameters sensitivity. In best fitting model, sensitivity positively correlated cloning efficiency media surface TAG-72 (a tumor-associated glycoprotein found epithelial ovary, lung, colon, breast origin) expression. If validated an vivo test such as nude mouse MTT could very useful new screening carcinoma, examining combination chemotherapy synergy, exploring strategies biochemical modulation, perhaps individualizing therapy when can established patient.
