摘要: Abstract Ten monoclonal antibodies to Alzheimer neurofibrillary tangles (ANTs) were produced by immunizing mice with a brain homogenate from senile dementia of the type (SDAT). In methanol-fixed isolated neuronal perikarya, six these reacted nearly every ANT, three recognized 70-88% ANTs, and one bound less than 30% ANT. paraffin sections, did not bind that had been fixed in formalin, stained weekly, four tissues formalin for more decade. Immunoblotting homogenates showed all but antibody proteins SDAT samples insoluble SDS too large enter even 3% polyacrylamide stacking gel. Polypeptides extractable Tris buffer molecular weight 58, 66, 70 kd detected both normal brains two only other antibodies. One show any reaction on immunoblot. The results demonstrate epitopes are identical ANTs contain unique antigenic determinants as well common brain. Whether acquired during tangle development or essential formation remains be investigated.
