摘要: Deviations from Hardy-Weinberg equilibrium (HWE) in control subjects may bias the estimates of genetic effects association studies (GAS) and meta-analysis. A large empirical evaluation was carried out to evaluate impact HWE deviation explore effect variance adjustment for allele-based odds ratio 833 individual GAS 72 meta-analyses. In GAS, any resulted stronger associations, 10 (1%) became significant (P < 0.05). One hundred sixteen (14%) showed 0.05); however, only 37 (4%) had more than 90% power detect at 5% level. meta-analyses, improved significance 53 meta-analyses (74%). Then, a formal statistical 0.05) revealed one previously negative whereas meta-analysis lost its significance. Between-study heterogeneity enhanced 50 (69%). None ( Q ) 0.10) meta-analysis, non significant. Sensitivity analysis not conforming applied 45 increased 26 (58%) seven (16%) were no longer Adjustment could be an effective strategy dealing with violations
