Genomic analysis of single cells from human basal cell cancer using laser-assisted capture microscopy

摘要: In this study, we show that direct mutational analysis of genomic DNA can be performed on single somatic cells extracted from a frozen, immunohistochemically stained tissue section using laser-assisted capture microscopy. Eighty-nine tumor were separately dissected one case human basal cell cancer (BCC) and p53 mutations analyzed by semi-automated sequencing PCR fragments. Amplification was obtained for at least the two exons approximately 50% cells. Identical found in widely spread areas tumor, suggesting clonal proliferation originating cell. Interestingly, comparison between results immunohistochemistry genetic revealed same irrespective immunoreactivity. We propose approach has great potential to allow investigation genotypic differences more specifically resolve important fundamental questions determining heterogeneity.
