摘要: The goal of the present study was to determine if expression metallothionein isoform 3 (MT-3) might serve as a biomarker for human bladder cancer. To accomplish this goal, we defined localization and MT-3 protein mRNA using fresh archival biopsy specimens obtained from patients undergoing differential diagnosis variety disorders. We used immunohistochemistry, immunoblot, RT-PCR analysis define mRNA. Immunohistochemical disclosed no immunoreactivity in normal cells. absence further confirmed by demonstrating that could not be detected reverse transcriptase-polymerase chain reaction (RT-PCR) or immunoblot. Immunohistochemistry also with interstitial cystitis related demonstrated expressed carcinoma situ (CIS), high-grade cancer, low-grade dysplastic lesions. immunostaining intense both CIS low moderate cancer determined subset these specimens; elevated compared housekeeping gene, ss-actin. cDNA primed contained FokI restriction site, site unique other MT family members. In conclusion, demonstrates is up-regulated up-regulation increases increasing tumor grade. finding minimal suggests developed into an effective
