摘要: Calcific aortic stenosis is the main heart valve disease in elderly, leading to massive focal calcification and thickening of cusps. Matrix metalloproteinases (MMPs) are thought contribute this process. Therefore, study assessed expression gelatinases MMP-2 MMP-9 endogenous tissue inhibitor metalloproteinase (TIMP)-2 as well gelatinolytic activity normal stenotic valves. Human tricuspid valves with without calcific were studied by immunohistochemistry for MMP-2, TIMP-2. The native sections was gelatin situ zymography or addition MMP activator p-aminophenymercuric acetate (APMA). Staining intensities TIMP-2 elevated compared controls. Minor staining present exclusively morphologic distribution comparable pattern since immunostaining demonstrated only a low number positive cells, observed likely due MMP-2. Gelatinolytic but significantly increased APMA.
