A developmental and tissue-specific enhancer in the mouse skeletal muscle acetylcholine receptor alpha-subunit gene regulated by myogenic factors.

摘要: The expression of the nicotinic acetylcholine receptor (AChR) in vertebrate striated muscle is regulated both during development and response to nerve-evoked activity. To define DNA sequences necessary for transcriptional regulation mouse alpha-subunit AChR gene, we have isolated analyzed alpha-gene 5'-flanking region. Primer extension RNase protection analysis showed that transcription initiates at 2 major 12 minor sites close translational initiation site. Using a series plasmids which segments region were linked bacterial chloramphenicol acetyltransferase (CAT) defined an 86-base pair enhancer sequence active C2 myotubes but not myoblasts or NIH3T3 fibroblasts. This contains three putative binding myoD1, 5'-upstream regions CAT transactivated by regulatory factors, myogenin. Transactivation MRF4 differed with specific construct tested. Whereas constructs containing homologous as well heterologous myosin light chain 1 promoter myoD1 myogenin, only their MRF4. Thus, gene information developmental specificity responsiveness myogenic factors.
