摘要: We addressed the question whether it is possible to lower threshold for naive T cells respond antigens. Purified adult and cord blood derived CD4+ CD45RA+ were incubated in presence of various cytokines two days (‘primed cells’), after which removed by extensive washing. Primed unprimed activated with solid phase-coupled anti-CD3 soluble anti-CD28 monoclonal antibodies (MoAb). Naive cells, primed interleukin(IL)-7 proliferated more vigorously than cells. required 6 h antigenic stimulation, whereas 20 h. The priming also shifted order stimulate antigen concentration a level. addition IL-10 almost completely abrogated enhancing effect IL-7 on Other (IL-1, IL-2, IL-6, IL-12, interferon (IFN)-γ, tumour necrosis factor (TNF)-α had less cell proliferation. However, no impact proliferation allogeneic immature or mature dendritic (DC). conclude that antigen-independent activation prior stimulation sensitizes may be help trying immune responses against weak This approach, however, does not enhance proliferative stimulated DC.
