Effects of recombinant human erythropoietin on megakaryocytes and on platelet production in the rat

摘要: The contention that erythropoietin (Epo) affects platelet production was investigated in the rat with recombinant human Epo (rHuEpo). In normal rats, caused a dose-dependent increase both reticulocyte and numbers, response preceding of platelets. Withdrawal resulted reticulocytes platelets returning to control levels. [75Se]-selenomethionine incorporation into also enhanced Epo. Chronic daily administration rHuEpo steady state erythrocyte levels after 12 14 days, which were elevated 20% above controls. Attainment this associated despite continued Epo, an effect not change half-life circulating polycythemic rats observed before on reticulocytes. Erythropoietin 2.4-fold frequency small acetylcholinesterase-positive cells within 24 hours, increased mean megakaryocyte diameter 48 hours. Furthermore, [3H]-thymidine labeling index megakaryocytes from treated for hours all stages maturation. These results support proposal causing production.
