摘要: We evaluated the antigenicity of recombinant infectious bronchitis virus (IBV) S1 protein expressed in mammalian cells. Recombinant was as a secreted fused with trimerization motif peptide, then purified using Ni Sepharose. The analyzed by Western blotting, mixed oil adjuvant, and administered to 29-day-old specific-pathogen-free chickens. Six weeks after immunization, anti-IBV neutralizing titer anti-S1 ELISA were determined; immunized chickens inoculated IBV via trachea ciliary activity observed. Results showed that highly glycosylated, lower than inactivated virus. However, indicated induced antibodies against S1. These results suggest may retain non-neutralizing epitopes but have unnatural glycosylation pattern conformation, resulting lacking conformational epitopes. In conclusion, from cells decreased, not sufficient induce antibodies.
