摘要: We used colloidal gold (1-nm diameter) with silver enhancement, in conjunction a low-temperature post-embedding immunolabeling technique, to localize several antigens normal skin at both the light and electron microscopic level within same tissue blocks. Normal subjected cyrofixation cryosubstitution embedded Lowicryl K11M was as substrate. Semi-thin sections (1 micron) were incubated primary antibody (against epidermal basement membrane zone associated two keratin sub-types), biotinylated secondary antibodies, then 1-nm gold-conjugated streptavidin. Finally, label enhanced using staining. Labeling of well demonstrated, area semi-thin showing best structural preservation greatest intensity identify part block be for ultra-thin sectioning. Ultra-thin treated similar procedure that employed sections. The labeling silver-enhanced probes intense readily visible by microscopy, even low magnification. have found this technique high degree specificity sensitivity intra- extracellular skin, added advantage providing means studies level.
