摘要: Differentiation and apoptosis are precisely regulated events in early embryogenesis. Retinoic acid-induced differentiation the embryonal carcinoma (EC) cell line NCR-G3 triggers concurrent induction of apoptosis. Using this system, which serves as a model embryogenesis, expression various bcl-2-related genes was analyzed these display either positive or negative regulatory effects on EAT/mcl-1, an antiapoptotic gene immediate gene, dramatically expressed at stage differentiation. Bcl-xL, another induced middle then gradually decreased to basal level. Expression Bax, proapoptotic molecule, detected high level remained relatively constant. Meanwhile, Bcl-2 Bcl-xS were below detectable levels throughout stages As balance bcl-2 is crucial step apoptosis, results suggest that EAT Bax likely regulate In later differentiation, down-regulation found coincide with gradual increase cells. Furthermore, use monoclonal antibody (3A2) specific revealed localized outer mitochondrial membrane human EC addition, immunoreactivity not apoptotic cells while it observed nearly all viable The findings rapid may prevent from undergoing thereby supporting viability
