Ultrasound Can Detect Macroscopically Undetectable Changes in Osteoarthritis Reflecting the Superficial Histological and Biochemical Degeneration: Ex Vivo Study of Rabbit and Human Cartilage

摘要: Recognizing subtle cartilage changes in the preclinical stage of osteoarthritis (OA) is essential for early diagnosis. To this end, ability ultrasound signal intensity to detect macroscopically undetectable change was investigated. In study, rabbit OA model and human samples examined by macroscopic evaluation, intensity, histology with Mankin scores, Fourier transform infrared imaging (FTIRI) analysis. Rabbit induced anterior cruciate ligament transection evaluated at 1, 2, 4 12 weeks. Twenty were harvested during total knee arthroplasty from patients who had normal (ICRS grade 0) on lateral femoral condyle. animal there no 2 weeks, but detected degenerative time point. Ultrasound also degeneration samples, all obtained intact site, however nearly (0≤ score <2), (2≤ <6), moderate (6≤ <10) actually intermixed. significantly different among these 3 stages well correlated scores (R = −0.80) FTIR parameters related collagen proteoglycan content superficial zone. conclusion, can microscopic deterioration when such do not exist macroscopically, reflecting histological biochemical changes.
