摘要: The objective of this study was to develop an efficient regeneration and transformation protocol for cucumber (Cucumis sativus L.). Cotyledonary node explants showed a very high frequency (94.3 %) when incubated on Murashige Skoog medium (MS) supplemented with 1.5 mg l−1 6-benzyladenine (BA). An Agrobacterium-mediated methodology developed using protocol. A construct containing sense mitogen-activated protein kinase gene (CsNMAPK) used transform cotyledonary nodes. Adventitious shoots were obtained in selection media 150 mg l−1 kanamycin 250 mg l−1 carbenicillin 4 weeks after bacterial inoculation. total twelve transgenic plants confirmed PCR-specific analysis. transgene copy number four (random selection) found be single Southern blot analysis the expression level CsNMAPK significantly higher than wild-type plants. rate approximately 4.8 %.
