摘要: For most pathogenic bacteria, the adherence to host cells is a crucial step for infection. Therefore, targeting bacterial attachment could be potential way treating infections. This thesis focuses on interactions between adhesin Multivalent Adhesion Molecule (MAM7) from commensal Escherichia coli strain HS with receptors, aiming understand molecular basis of these interactions. Targeting MAM7 and was one this study’s aims towards developing anti-bacterial agents. In study, HSMAM7 based adhesion inhibitors- beads coupled an engineered bacterium expressing HSMAM7- were designed their role as anti–adhesion agents against bacteria investigated. study demonstrates that inhibitors have ability displace cells. Dissecting binding receptors second part study. The findings revealed binds sulfatide, several components extracellular matrix (ECM) mucin. Further investigation interaction HSMAM mucin lipid sulfatides via specific recognition shared 3-O-sulfo-galactosyl moiety in sulfation dependent manner. Sulfatase-producing Bacteroides thetaiotaomicron affect inhibited presence B. means E. would free able attach epithelial cells, finding relevance inflammatory bowel diseases (IBDs).
