摘要: In rheumatoid arthritis (RA) the proliferation of synovial lining cells appears to be one initial pathologic changes that contributes destruction articular joints. To understand pathomechanisms involved in these functional changes, we analyzed transcriptional regulation zinc-finger gene 225 (Z-225/Egr-1), a transcription factor expressed immediate early events cellular activation. We found Z-225 transcripts were significantly up-regulated RA synoviocytes. primary and long term culture was spontaneously transcribed at elevated levels. situ hybridization probe showed characteristic tissues. Identity signals product confirmed freshly isolated tissue by enzymatic amplification cDNA PCR technique. also detected characterized library established from explant. therefore conclude synoviocytes produce products Because growth response is expression other genes such as proto-oncogenes c-ras c-sis, which are synovium, activation may represent key event joint destruction.
