摘要: Biosynthesis of xanthan polysaccharide, a virulence factor phytopathogenic Xanthomonas campestris pv. (Xcc), involves the gum operon and cyclic AMP receptor protein (CRP) homologue Clp. Clp was shown to have same DNA binding specificity as CRP at positions 5, 6, 7 (GTG motif) left arm. Therefore, sites (CBSs) typically been identified by pattern searching Xcc genome using consensus sequence. Here, results reporter assay electrophoretic mobility shift suggest that upregulates two non-consensus sites, in which more conserved right arm may compensate for lack conservation arm, high GC content central region (6 bp) be important binding, enhanced if GC-rich is palindromic. These atypical CBSs exist promoters Clp, while retaining capacity bind typical CBSs, has evolved CBS because: 1) shares only moderate homology with modulated di-GMP; 2) higher (65%) than Escherichia coli (50%).
