Experimental autoimmune dacryoadenitis: purification and characterization of a lacrimal gland antigen.

摘要: A soluble lacrimal gland antigen (LG-Ag) capable of inducing experimental autoimmune dacryoadenitis (EAD) in SJL/J mice has been purified from bovine extracts by a combination ion exchange and gel-filtration chromatography. Sodium dodecyl sulfate polyacrylamide gel electrophoresis showed single band with mobility corresponding to molecular weight 45,000 daltons. Antibodies raised guinea pigs against LG-Ag were used confirm the tissue-specificity assess cellular localization within tissues. Anti-LG-Ag serum reacted crude but not submaxillary gland, parotid or retina. As shown immunofluorescent staining, appeared be localized on murine ductal epithelial cells was lacking salivary ducts. induced EAD that characterized infiltration mononuclear around ducts associated vasculature, accompanied extensive damage acinar cells. The lesions tissue-specific, immunized had intense lymphocytic infiltrates no significant their harderian glands. availability LG can facilitate further analysis pathogenetic mechanisms EAD.
