摘要: Dry eye disease is a very common condition that causes morbidity and healthcare burden decreases the quality of life. There need for suitable dry animal model to test novel therapeutics treat autoimmune conditions. This protocol describes chronic rat model. Lewis rats were immunized with an emulsion containing lacrimal gland extract, ovalbumin, complete Freund's adjuvant. A second immunization same antigens in incomplete adjuvant was administered two weeks later. These immunizations subcutaneously at base tail. To boost immune response ocular surface glands, extract ovalbumin injected into forniceal subconjunctiva glands 6 after first immunization. The developed features, including reduced tear production, decreased stability, increased corneal damage. Immune profiling by flow cytometry showed preponderance CD3+ effector memory T cells eyeball.
