Genetic diversity and relationships of sweetpotato and its wild relatives in Ipomoea series Batatas (Convolvulaceae) as revealed by inter-simple sequence repeat (ISSR) and restriction analysis of chloroplast DNA

摘要: Genetic diversity and relationships of 40 accessions Ipomoea, representing ten species series Batatas, were examined using ISSR markers restriction-site variation in four non-coding regions chloroplast DNA. A total 2071 fragments generated with 15 primers these and, on average, 52 bands per accession amplified. Most the contained dinucleotide repeats. The highly polymorphic (62.2%) among studied. Restriction analysis (cp) DNA revealed 47 informative length mutations. Phylogenetic analyses cpDNA datasets generally similar at interspecific level, but high polymorphism ISSRs resulted a better separation intraspecific accessions. However, combined dataset appeared to be appropriate resolving both intra- relationships. Of examined, I. trifida was found most closely related cultivated sweetpotato, hexaploid I.batatas, while ramosissima umbraticola distantly batatas within series. Ipomoea triloba, hitherto considered one ancestors only sweetpotato based similarity index.
