摘要: MicroRNAs (miRNAs) are important regulatory molecules in most eukaryotes and identification of their target mRNAs is essential for functional analysis. Whereas conventional methods rely on computational prediction subsequent experimental validation RNAs, we directly sequenced >28,000,000 signatures from the 5' ends polyadenylated products miRNA-mediated mRNA decay, isolated inflorescence tissue Arabidopsis thaliana, to discover novel miRNA-target RNA pairs. Within set approximately 27,000 transcripts included 8,000,000 nonredundant signatures, several previously predicted but nonvalidated targets miRNAs were found. Like validated targets, showed a single abundant signature at miRNA cleavage site, particularly libraries mutant deficient 5'-to-3' exonuclease AtXRN4.
