Electron microscopic examination of ribosome preparations from germinated spores of Aspergillus fumigatus.

摘要: Ribosomes were isolated from germinated spores (germlings) of Aspergillus fumigatus and electron microscopy was used to determine qualitatively the extent cellular contamination. After differential centrifugation, initial crude preparation contained 80S ribosomes numerous membrane contaminants in form sheets, palisade aggregates, vesicles 0.10–0.18 μm size. Gel filtration chromatography Sepharose CL-4B did not remove all Homogenous ribosome suspensions, morphologically free other component contamination, then obtained after gel fractions centrifuged through a 35% sucrose solution. The total yield germling approximately four times comparable spore preparation. diffusion precipitin patterns identical those with both antisera.
