摘要: BACKGROUND Solid tumors are angiogenesis dependent, and elevated levels of proangiogenic cytokines have been reported in a variety histologies. Endostatin is an antiangiogenic fragment the basement membrane protein, collagen XVIII. Because protein fragments may be generated by tumor-derived proteases, authors sought to determine whether circulating endostatin were patients with localized soft tissue sarcoma. METHODS The analyzed preoperative serum endostatin, vascular endothelial growth factor (VEGF), basic fibroblast (bFGF) 25 (14 males 11 females; mean age, 44 years) sarcoma. For each sample, two aliquots assayed duplicate using competitive enzyme immunoassay. Serum compared from 34 age-matched gender-matched volunteer blood donors. RESULTS Endostatin significantly higher sera sarcoma than healthy controls (43.0 ng/mL vs. 25.8 ng/mL, respectively; P = 0.0002; Mann–Whitney U test). Significant elevations also noted VEGF bFGF (P 0.0002 0.0001, respectively). Furthermore, > 2 standard deviations above control (55 ng/mL) associated increased risk tumor recurrence after resection 0.047; log-rank test). CONCLUSIONS Serum VEGF, Elevated appear aggressiveness. The role these as potential targets for therapy warrants further study. Cancer 2001;91:1525–9. © 2001 American Society.
