摘要: Sterol carrier protein-2 (SCP-2) is an intracellular protein of Mr 13,096. In vitro studies have shown that it involved in the transport and metabolism cholesterol. This believed to participate these activities by forming a stoichiometric complex with sterol. Because occur different locations, i.e. mitochondria, peroxisomes, cytosol, can be predicted SCP-2 targets sites. this report we show mouse cDNA (785 base pairs) encodes precursor form containing N-terminal presequence additional C-terminal residue. These amino acid residues are found proteins targeted mitochondria respectively. signals not purified from rat liver cytosol which cytosolic form. Northern analysis shows there four species mRNA hybridize SCP-2-specific probe at 1.0, 1.7, 2.2, 2.9 kilobases. Southern gene distributed over large amount DNA or multiple genes. We cloned cytosolic/peroxisomal into Escherichia coli expression vector pKK233-2 expressed recombinant SCP-2, 13,034. The immunoreactive rabbit anti-rat antibody. It also has biological activity equivalent homogeneous stimulating microsomal conversion 7-dehydrocholesterol cholesterol esterification acyl-CoA acyltransferase microsomes.
