摘要: Formation of the autophagosome is likely most complex step macroautophagy, and indeed it morphological functional hallmark this process; accordingly, critical to understand corresponding molecular mechanism. Atg8 only known autophagy-related (Atg) protein required for formation that remains associated with completed sequestering vesicle. Approximately one-fourth all characterized Atg proteins participate in biogenesis affect Atg8, regulating its conjugation phosphatidylethanolamine (PE), localization phagophore assembly site and/or subsequent deconjugation. An unanswered question field regards physiological role deconjugation Atg8-PE. Using an mutant bypasses initial Atg4-dependent processing, we demonstrate important facilitate multiple events during macroautophagy. The inability deconjugate Atg8-PE results mislocalization vacuolar membrane. We also show efficient biogenesis, Atg9-containing tubulovesicular clusters into phagophores/autophagosomes, disassembly PAS-associated components.
